This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate of the kit has been pre-coated with an antibody specific to Human ROMO1. Test samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a detection antibody (biotinylated) specific for Human ROMO1 and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each well of the well and then the plate is incubated for specific time. The free conjugate is washed away by buffer. The substrate solution is added to each well. The well that contains Human ROMO1, detection antibody (biotinylated) and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is completed by the addition of stop solution and the color turns yellow. In the last, the optical density (OD) of the wells are measured by spectrophotometer at a wavelength of 450 nm ? 2 nm. The OD value comes in proportional to the concentration of Human ROMO1. The concentration is calculated for Human ROMO1 in the samples by comparing the OD of the samples with the standard curve.
