Western Blot is laboratory method to separate and analyzes any specific protein in the sample containing mixture of different proteins. The method is used in Cell and Molecular Biology analysis of proteins. The overview of the western blot method include
1) Processing of protein samples for analysis,
2) Separation of size specific proteins by electrophoreses,
3) Transfer of proteins bands on nitrocellulose membrane,
4) determining the proteins with the help of protein specific primary antibodies and secondary antibodies.
The term blotting refers to transfer of biological samples from a gel of separation to a solid platform such as nitrocellulose membrane and determination or identification of proteins. Western Blotting is also known as protein blotting or immunoblotting. The method is based on the principle of immunochromatography in which proteins are separated in polyacrylamide gel on the basis of their size i.e molecular weight. The SDS-PAGE technique is best suitable for western blotting. The method provides qualitative or semi quantitative results.
